M13 mp18 RF DNA

Description:

This DNA is a phage vector suitable for DNA sequencing by dideoxy method. Since it has multi-cloning sites on lacZ' region, any DNA insertion into this phage vector can be easily verified using soft-agar plates containing IPTG and X-Gal. Moreover, the cloned DNA can be directly used for DNA sequencing, because it is a single-strand DNA when isolated from the phage particle. M13 primer is available for DNA sequencing. Using Deletion Kit for Kilo-Sequencing (Cat.# TAK 6030), DNA sequencing of the cloned DNA with kilo-bases can also be performed.

Notes:

When cloning for a long DNA fragment (e.g. above 4 to 5 kbp) using M13 phage vector, some deletion is often found during the culture.

Purity

  • Contains over 70% double-strand covalently closed circular form I (RF I).
  • It is confirmed that the multi-cloning site is well-maintained upon sequence analysis by dideoxy method.
  • It is confirmed that the vector contains a single cleavage site for EcoR I, Sac I, Kpn I, Sma I, BamH I, Xba I, Sal I, Pst I, Sph I and Hind III upon restriction analysis by the enzymes.
